摘要:
通过微波消解法,原子吸收分光光度计对当归中铅的吸光度进行测定,采用Box-Behnken Design设计试验方法以确定最佳仪器分析条件;通过单因素试验,选取灰化温度、灰化时间、原子化温度、原子化时间四因素进行三水平试验,通过Box-Behnken Design建立数学模型,方差分析、响应曲面分析、愿望函数优化寻求最佳条件;结果显示建立的数学模型R2=0.9671,R2Adj =0.9342说明该模型具有很好的可信度,可用此模型进行预测,变异系数 CV %=3.62 %,表明在该试验条件下,变异程度较小,该模型具有较好的重复性,当灰化温度为525℃,灰化时间为20s,原子化温度为1550℃,原子化时间为5s时吸光度最大为0.24;因此,本研究可为原子吸收分光光度计测定铅元素提供很好的方法理论基础。

Abstract:
The absorbance of lead in flour was determined by microwave digestion method and atomic absorption spectrophotometer. Box-behnken Design was used to determine the best instrumental analysis conditions. Through the single-factor test, the four factors of ashing temperature, ashing time, atomization temperature and atomization time were selected for the three-level test. The mathematical model was established by Box-Behnken Design, and the optimal conditions were sought by variance analysis, response surface analysis and desire function optimization. Results show that the established mathematical model of the R2=0.9671,R2Adj =0.9342 shows that the model has good credibility, this model could be used to predict the coefficient of variation CV % = 3.08%, indicates that under the condition of the experiment, variation is small, the model has good repeatability, when ashing temperature is 525℃, ashing time is 20s, atomization temperature is 1550℃, atomization time for 5s absorbance maximum 0.24; Therefore, this study can provide a good theoretical basis for the determination of lead by atomic absorption spectrophotometer.
